Milton A. Wolf was born in Cleveland, Ohio on May 29, 1924, the son of Sam and Sylvia (Davis) Wolf. He graduated from Glenville High School in 1942 and served in the Army Air Corps during World War II. After the war, Wolf attended The Ohio State University and graduated with a B.A. in Chemistry and Biology in 1948.
